γ-MSH — gamma-melanocyte-stimulating hormone — is the melanocortin that refuses to fit the pattern. Most peptides in this family, led by α-MSH, are broad-spectrum signals that touch several receptors and headline in pigment and appetite. γ-MSH is comparatively picky about its receptor, and its main claim to fame is somewhere else entirely: the control of sodium and blood pressure.[2]

Where α-MSH activates four of the five receptors strongly, γ-MSH is relatively MC3R-preferring — it is the natural melanocortin best able to engage MC3R at physiological concentrations, with only weaker activity at MC1R, MC4R, and MC5R.[3] That selectivity is exactly why MC3R and γ-MSH tend to be discussed as a pair: the receptor's quieter, harder-to-pin-down physiology and its most selective ligand belong to the same story.

γ-MSH's distinctive role is in sodium balance. It is a natriuretic signal — it promotes the excretion of sodium — and appears to participate in the physiological regulation of the body's salt handling, an effect linked to MC3R expressed in the kidney.[1] At the same time, acting through central nervous system pathways, γ-MSH can raise blood pressure by stimulating sympathetic outflow.[2] The result is a melanocortin whose physiology reads like cardiovascular and renal biology rather than skin or appetite.
